FGF23 inhibitor 13a (FGF23-IN-13a) is a ZINC13407541 analogue with enhanced drug-like properties, inhibits FGF23 with IC50 of 0.14 uM.
FGF23 inhibitor 13a exhibits greater effects to increase Npt2a and Npt2c message expression, consistent with higher serum phosphate levels in kidney of Hyp mice.
FGF23 inhibitor 13a demonstrates short- and long-term efficacy in blocking FGF23 end-organ effects in Hyp mice, a human homologue of XLH. FGF23 inhibitor 13a efficiently abrogates aberrant FGF23 signaling, corrects hypophosphatemia and aberrant 1,25(OH)2D levels, and improves both bone and renal gene expression profiles caused by elevated FGF23 levels in Hyp mice.
