Eltrombopag (SB497115, DDL-701) is a nonpeptide, orally active thrombopoietin receptor (TpoR) agonist with EC50 of 0.27 uM in cell-based assays, also inhibits the SARS Co-V-2 papain-like protease (PLpro) with IC50 of 13 uM.
Eltrombopag is specific to the human and chimpanzee TpoR, activates the signaling transducers and activators of transcription (STAT) and mitogen-activated protein kinase signal transduction pathways.
Eltrombopag stimulated BAF3/hTpoR cell proliferation with EC50 of 0.03 uM, as well as the differentiation of BAF3/hTpoR cells.
Eltrombopag interacts selectively with the TpoR without competing with Tpo, leading to the increased proliferation and differentiation of human bone marrow progenitor cells into megakaryocytes and increased platelet production.
