ER-000444793 is a potent, cyclophilin D-independent inhibitor of mitochondrial permeability transition pore (mPTP) with with IC50 of 2.8 uM.
ER-000444793 neither affects cyclophilin D (CypD) enzymatic activity, nor displaces CsA from CypD protein.
ER-000444793 improves mitochondrial Ca2+ tolerance with IC50 of 1.2 uM, increases mitochondrial Ca2+ retention capacity dose-dependently, and delays the onset of mPT in HeLa S3 cells with no effect on mitochondrial or cellular health.
