DW18134 is a potent, selective IRAK4 kinase inhibitor with IC50 of 11.2 nM, inhibits the IRAK4-induced activation of NF-κB signaling.
DW18134 inhibited TNF-α and IL-6 secretion in a dose-dependent manner in PCM and RAW264.7 cells, did not affect the proliferation of macrophages.
DW18134 (15 mg/kg and 30 mg/kg, i.p.) ameliorated inflammatory phenotype and acute tissue damage in LPS-induced peritonitis mice.
DW18134 reduced macrophage infiltration and pro-inflammatory gene expression and abrogated IRAK4 signaling pathway activation in LPS-induced peritonitis mice.
DW18134 (2 mg/kg, i.p.) ameliorated symptoms and reduced intestinal pathological damage in mice with DSS-induced experimental colitis.
