DSK638 (DSK-638) is a potent small moelcule RECK inducer, binds histone deacetylases (HDACs) with IC50 of 5-10 uM for HDAC1/3.
DSK638 exhibits bioactivities similar to RECK, such as induction of flat reversion in v-K-RAS-transformed cell, suppression of Matrigel-invasion by HT1080 cells and inhibition of pro-MMP2 activation.
DSK638 inhibits spontaneous lung metastasis in our mouse xenograft model. Induction of RECK expression involves SP1 sites in its promoter and may be mediated by KLF2.
DSK638 also upregulates MXI1, an endogenous MYC-antagonist, and inhibition of metastasis by DSK638 is dependent on both RECK and MXI1.
