DO264 (DO-264) is a potent, selective, reversible and vivo-active inhibitor of ABHD12 that inhibits ABHD12-dependent lyso-PS lipase activity of membrane lysates from mouse brain with IC50 of 2.8 nM.
DO264 blocks lyso-PS hydrolysis activities of recombinant mouse and human ABHD12 in transfected HEK293T cell lysates with IC50 of 30-90 nM.
DO264 selectively blocks >95% of ABHD12 activity without affecting the activity of other serine hydrolases in THP-1 cells at 1 uM.
DO264 elevates lyso-PS in mouse brain and primary human macrophages, inhibits ABHD12 and increases brain lyso-PS content, stimulates immune responses in vivo.
