DNMT/HDACi 15a is a potent, dual DNMT and HDAC inhibitor with IC50 of 57, 17 nM for HDAC1, HDAC6 respectively.
DNMT/HDACi 15a exhibits greater potency in inhibiting DNMT1 activity than DNMT3A and DNMT3B.
DNMT/HDACi 15a regulates the activities of DNMT1 and HDAC1 in breast cancer, decreases global DNA demethylation, induces downregulation of DNMT1 protein expression and upregulation of H3 acetylation in MDA-MB-453 and BT-474 cells.
DNMT/HDACi 15a exerts antitumour effects in vitro and in vivo, promote apoptosis and induce cell cycle arrest.
