DCEM1 is a mitochondrial unfolded protein response (UPRmt) inhibitor, specifically disrupts HSP60-ClpP interaction in PCa cells and in vitro, suppresses prostate cancer growth in vivo.
DCEM1 binds to the apical domain of HSP60.
DCEM1 did not alter the protein levels of UPRmt components including HSP60, HSP10, ClpP and LONP1 in most of PCa cells and normal prostate epithelial cells, and in human embryonic HEK-293 cells.
DCEM1 robustly downregulated c-Myc and EZH2, and reduced AR and inhibited AR activity in PCa cells.
DCEM1 induced metabolic stress in PCa cells, downregulated c-Myc, EZH2 and AR as well as inhibits tumor growth in vivo.
