D-583 is a potent hybrid D2/D3 agonist and iron (II)-specific chelator with EC50 of 3.14 nM (Emax=63%) and 0.62 nM (Emax=68%) for D2 and D3 receptors respectively, exhibits anti-ferroptotic activity.
D-583 (0.1-20 uM) significantly protect PC12 cells from the toxic effects of 6-OHDA.
D-583 (0.1-20 uM) dose dependently protect PANC-1 cells from the toxicity of RSL3.
D-583 (1 uM, 10 uM) induces dose dependent reduction of ROS in PANC1 cells.
D-583 dose dependently restored ferroptosis marker proteins GPX4, SLC7A11 and Nrf2.
D-583 (5 µMol/kg)s ignificantly reversed akinesia in Parkinson's disease animal model.
