Cl-NIO is a potent, selective inhibitor of human dimethylarginine dimethylaminohydrolase-1 (hDDAH-1) with Ki of 1.3 uM, > 500-fold selectivity against arginase and endothelial nitric oxide synthase.
Cl-NIO displays cellular inhibition of DDAH-1 artificially expressed within cultured HEK293T cells with IC50 of 6.6 uM.
Cl-NIO inhibits DDAH-1 within cultured human cells and leads to downstream inhibition of nitric oxide biosynthesis.
Cl-NIO treatment (10 uM) resulted in a concentration-dependent decrease in NO2− + NO3− levels in A375 melanoma cell line, which reflects decreased nitric oxide production.
Cl-NIO is a suitable tool for studies of methylarginine-mediated regulation of nitric oxide production and as a potential therapeutic lead for a variety of indications with elevated nitric oxide levels.
