CX4338 (CX-4338) is a potent, selective CXCR2 inhibitor that selectively inhibits CXCR2-mediated recruitment of β-arrestin-2 with IC50 of 6.3 uM.
CX4338 shows little to no inhibition of CXCR4-mediated β-arrestin-2 recruitment at 100 uM.
CX4338 inhibits receptor internalization, while enhancing CXCR2-mediated MAPK activation.
CX4338 inhibits CXCL8-induced chemotaxis in CXCR2-overexpressing cells and human neutrophils.
CX4338 significantly reduces neutrophils in bronchoalveolar lavage induced by LPS in mice.
