CVB4-57 is a specific, reversible small molecule Connexin26 hemichannel (Cx26 hemichannel), reversibly inhibited the dye uptake at 5 uM without affecting the gap junction communication and barrier function.
CVB4-57 (1 uM) decreases the dye uptake in cells expressing hCx26 to 79 ± 17%.
CVB4-57 (20 uM) decreases the dye uptake in N2A cells expressing Cx46 to 72 ± 3%.
CVB4-57 could block Cx hemichannels with minimal impact on the epithelial physiology.
Cx26 hemichannels are more sensitive to both agents compared to Cx46.
