CNS-11g is a small molecules capable of disassembling preformed alpha-synuclein fibrils, may exert it's disaggregating effects on the N terminus of the fibril core.
CNS-11g is more predictive of oral bioavailability and central nervous system penetration compared to EGCG.
CNS-11g (10 uM) inhibits intracellular seeding in alpha-synuclein biosensor cells and mitigation of alpha-synuclein cytotoxicity.
CNS-11g disaggregates MSA patient brain-derived alpha-synuclein fibrils in vitro, reduces alpha-synuclein aggregates in C. elegans, also could penetrate brain tissue in mice.
