CNI103 (CNI-103) is a highly potent, cell-permeable, metabolically stable peptidyl inhibitor of Calcineurin-NFAT interaction wioth KD 16 nM and IC50 of 220 nM.
CNI103 selectively blocks the interaction between calcineurin and NFATc3, specifically inhibits calcineurin signaling in vitro and in vivo.
CNI103 exhibits a favorable pharmacokinetic profile, broad tissue distribution following different routes of administration, and minimal toxicity.
CNI103 significantly reduces TNFα and IL-6 secretiona gainst LPS-induced ALI in mice.
CNI103 is a promising novel treatment for Acute respiratory distress syndrome (ARDS) and, possibly, COVID-19 ARDS and other inflammatory diseases.
