Brusatol (NSC 172924, (+)-Brusatol) is a quassinoid extracted from Brucea Esters, can inhibit HIF-1 signaling pathway, also is a potent Nrf2 inhibitor.
Brusatol selectively reduces the protein level of Nrf2 through enhanced ubiquitination and degradation of Nrf2.
Brusatol enhances the efficacy of cisplatin, induces apoptosis, reduces cell proliferation, and inhibits tumor growth more substantially when compared with cisplatin treatment alone in A549 xenografts.
Brusatol down-regulates HIF-1α protein levels under hypoxia or CoCl2-induced mimic hypoxia in HCT116 cells without causing significant cytotoxicity.
