Bcl3 inhibitor JS6 is a novel potent, antimetastatic Bcl3 inhibitor, targeting the Bcl3-p50 interaction site.
Bcl3 inhibitor JS6 suppresses NF-kB signaling in NF-kB luciferase reporter assay and inhibits multimodal migration of breast cancer cells with IC50 of 310 nM.
JS6 treatment led to reductions in Bcl3-NF-kB1 binding, tumor colony formation, and cancer cell migration in vitro. JS6 inhibited tumor stasis and showed antimetastatic activity in vivo, while being devoid of overt systemic toxicity.
