BT503 is a potent small molecule agonist of Krüppel-Like Factor 15 (KLF15) with EC50 of 7.1 nM in luciferase reporter assays, directly inhibits NF-κB signaling by targeting IKKβ (IC50=163 nM).
BT503 induces KLF15 activity in pEZX FR03hKLF5p-mutant podocytes, induces KLF15 activity independent of glucocorticoid signaling.
BT503 improves cell viability in podocytes under cell stress.
BT503 demonstrated protective effects in cultured human podocytes and in three independent proteinuric models (lipopolysaccharide, nephrotoxic serum nephritis, HIV-1 transgenic mice).
BT503 directly inhibits IKKβ from phosphorylating IκBα, NF-κB inhibitory subunit, which prevented the nuclear translocation of NF-κB dimers, and, in turn, restores KLF15 levels under podocyte stress.
