BI 6015 (BI-6015) is a potent synthetic HNF4α antagonist, binds to HNF4α with high affinity and modulates HNF4α target genes and metabolic processes controlled by HNF4α.
BI 6015 reduced endogenous insulin gene expression 50-fold in T6PNE cells, and strongly repressed HNF4α gene expression in MIN6 and HepG2 cells.
BI 6015 induced steatosis in primary mouse hepatocytes, again without evidence of cell death in vitro, and induced loss of HNF4α expression and hepatic steatosis in vivo.
BI 6015 is also cytotoxic to human hepatocellular carcinoma (HCC).
