BC-K-YH16899 (YH16899) is a specific small molecule inhibitor of prometastatic lysyl-tRNA synthetase-laminin receptor interaction (KRS-67LR PPI), suppresses cancer metastasis.
YH16899 specifically blocks the membrane function of KRS with little influence on the catalytic activity of KRS.
YH16899 treatment decreased the amount of membranous 67LR, but not that of cytosolic 37LRP in in vitro pull-down and IP assays. YH16899 inhibited H226 migration (high level of 67LR) with IC50 of 8.5 uM, inhibited migration of cancer cells more significantly than that of normal cells.
YH16899 suppressed metastasis in metastasis model of invasive A549 cells, without severe toxicity.
YH16899 suppressed membrane translocation of KRS, showed no effect on KRS binding to AIMP2.
