BC-1293 is a specific small molecule inhibitor of bacterial-induced ubiquitin E3 ligase subunit FBXO24, abrogates FBXO24-DARS2 binding, exhibits immunostimulatory properties.
BC-1293 potentiated Pam3CSK4 induced secretion of both endogenous and ectopically expressed Flag-DARS2 from cells.
BC-1293 significantly increased secretion of IL-6, TNFα, and IL-1β from BEAS-2B, CD14 + , and THP-1 cells respectively, compared to vehicle or Pam3CSK4 alone.
BC-1293 also triggered release of TNFα and IL-1β from BEAS2-2B and CD14+ cells, respectively
BC-1293 (0, 1, 10 and 20 μM) displayed a significant stimulatory effect of BC-1293 on IL-6 secretion in Fbxo24-WT precision cut lung slice (PCLS) cultures, but not PCLS derived from Fbxo24+/- mice.
