BAX inhibitor BAI1 (BAI1) is a small-molecule allosteric inhibitor of BAX, bind directly to BAX and inhibits tBID-induced BAX-mediated membrane permeabilization with IC50 of 3.4 uM.
BAI1 binds to inactive monomeric BAX at a novel allosteric site, allosterically inhibits BIM BH3-mediated BAX activation and oligomerization.
BAI1 selectively inhibited BAX-mediated apoptotic cell death, effectively inhibited TNFα + cyclohexamide mediated-apoptosis monitored by caspase 3/7 activation in a dose-dependent manner (IC50=1.8 uM).
