Autogramin-1 is a potent autophagy inhibitor, inhibits autophagy induced by either starvation or mTORC1 inhibition with IC50 of 0.17 and 0.44 uM, respectively.
Autogramin-1 also inhibits LC3 lipidation and the degradation of p62, and selectively inhibits growth of glucose-starved MCF7 cells as compared to fed cells.
Autogramin-1 selectively targets cholesterol transfer protein GRAM domain-containing protein 1A (GRAMD1A) and directly competes with cholesterol binding to the GRAMD1A StART domain.
GRAMD1A is required for autophagosome initiation.
