AZ32 (ATM inhibitor AZ32) is a potent, selective, orally bioavailable, BBB-penetrating ATM inhibitor with IC50 of <6.2 nM, effectively blocks ATM auto-phosphorylation at S1981 with IC50 of 0.31 uM.
AZ32 shows radiosensitizing and DDR blocking ability in vitro in a panel of human glioma cell lines with different p53 status, including LN18, U1242, and T98G (all with mutant p53), as well as DBTRG and U87MG (with wild-type p53).
AZ32 is the first ATMi with oral bioavailability shown to radiosensitize glioma and improve survival in orthotopic mouse models.
