AZ19-3-2 is a high affinity, dual-head inhibitor of carbonic anhydrase IX (CA IX) with binding Kd of 25 pM in thermal shift assays, binds two CA IX catalytic domains on the cell surface simultaneously.
AZ19-3-2 possesses an extremely high affinity for CA IX, exhibits the Kd between 10 and 100 nM (CA II, CA VB, CA VI, CA XII, CA XIII and CA XIV) with 1000-fold selectivity.
AZ19-3-2 effectively mitigates the acidification induced by CA IX and reduces spheroid growth under hypoxic conditions in the SK-N-AS cell line.
AZ19-3-2 also diminishes the secretion of pro-tumour chemokines IL-8 (CXCL2) and CCL2.
AZ19-3-2 combined with CCR2 antagonist inhibits tumour growth in mice SK-N-AS xenograft.
