AZ137 is a potent and selective Homeodomain-interacting protein kinase 4 (HIPK4) inhibitor with biochemical IC50 of 11 nM and cellular EC50 of 76 nM.
AZ137 displays over 100-fold selectivity over HIPK1-3, also exhibits excellent kinome-wide selectivity in scanMAX™ panel (KINOMEscan®, Eurofins DiscoverX).
AZ137 (1-5 uM) demonstrates pharmacological inhibition of HIPK4, blocks F-actin remodeling in NIH-3T3 cells, disrupts actin architecture, returning it to a normal fibrous structure.
