AZ12216052 is potent, selective positive allosteric modulator of mGluR8, potentiates the effect of 300 nM glutamate with EC50 of 1.0 uM in [35S]GTPγS binding assays.
AZ12216052 shows no effect in GTPγS binding assays using membranes prepared from mGluR4-expressing GHEK cells.
The positive allosteric modulator effects of AZ12216052 were inhibited by the group-II/III antagonists LY341495 and CPPG.
