ARN1468 is an anti-prion, cellular active small molecule targeting SERPINA3 (ITC KD=26 uM), decreases prion accumulation and reduces PrPSc in the ScGT1 RML cell line with EC50 of 8.64 uM.
ARN1468 exhibits anti-prion effect in ScGT1 22L, ScN2a RML, and ScN2a 22L cell lines, reduces PrPSc levels by 35% on average, reduces prion levels by almost 85% in ScN2a cells infected with 22L prion.
Anti-prion activity of ARN1468 does not affect PrP petabolism, localization, or aggregation propensity.
ARN1468 and quinacrine act synergically to clear prions in ScN2a RML cells.
