ALLO-2 is a potent small molecule Smoothened (Smo) antagonist that inhibits Smo agonist Hh-Ag1.5-induced luciferase expression in TM3-Gli-Luc cells with IC50 of 6 nM.
ALLO-2 competes with [3H]cyclopamine with an IC50 of 2.8 nM.
ALLO-2 binds to Smo differently than GDC-0449 and other Smo antagonists, and inhibits both wild-type and D473H mutant human SMO with similar potencies.
