ALKBH5 inhibitor TD19 is a specific, covalent inhibitor of RNA demethylase ALKBH5, inhibits ALKBH5-induced demethylation with IC50 of 1.5-3 uM.
TD19 does not inhibit FTO and ALKBH3 at 100 uM. Displays Kd value of 145 nM in isothermal titration calorimetry (ITC) assay, and Kd of 17.5 nM in microscale thermophoresis (MST) assays.
TD19 irreversibly modifies the residues C100 and C267, inhibits ALKBH5 binding to m6A-RNA substrate.
TD19 displays good anticancer efficacy in acute myeloid leukemia and glioblastoma multiforme cell lines (MOLM13 cell IC50=9.5 uM, U87 cell IC50=7.2 uM).
