ALDH1A1 inhibitor N42 (FSI-TN42) is a potent, specific and orally bioavailable ALDH1A1 inhibitor with IC50 of 23 nM, 800-fold selectivity over ALDH1A2 (IC50=18 uM).
N42 strongly and irreversibly binds to ALDH1A1.
N42 (10-100 mg/kg) causes retinoic acid (RA) synthesis inhibition in vivo.
N42 affects retinoid storage and metabolism in a tissue-dependent manner, and protects against diet-induced obesity in vivo.
