AFN700 (AFN 700) is a specific inhibitor of IκKβ kinase with IC50 of 0.51 uM in NF-kB reporter gene assays, inhibits TNF-a secretion produced by THP-1 cells (3h treatment with 100 ng/ml Pam3CSK4) with IC50 of 0.89 uM.
AFN700 also efficiently blocked IL-1b release triggered by nigericin in THP-1 cells with IC50 of 0.29 uM.
AFN700 blocked NLRP3-driven IL-1b production independently of NF-kB, blunted the competence of the NLRP3 inflammasome complex.
