AES-135 (AES135) is a novel small molecule HDAC inhibitor, inhibits HDAC3, HDAC6, HDAC11 with IC50 of 654, 190, 636 nM, with low-uM activity against HDAC8 (IC50=1.2 uM).
AES-135 exhibits selective in vitro cytotoxicity (IC50=1-4 uM) in low-passage patient-derived pancreatic cancer cells even in the presence of cancer-associated fibroblast (CAF) cells.
AES-135 has favorable in vivo properties such as metabolic stability in mouse hepatocytes and bioavailability in micromolar (μM) concentrations in NSG mice for >10 h (IP injection).
AES-135 increased survival of C57Bl/6 mice implanted with KPC tumor cells in vivo.
