AE-848 is a small molecule modulator of NF-κB and PI3K/AKT/mTOR signaling pathways, shows potent in vitro and in vivo anti-myeloma activity.
AE-848 (5 µM, 24 and 48 h) significantly induces apoptosis of U266 and RPMI8226 cells, markedly induces loss of MMP.
AE-848 induces cleavage of caspase-8, caspase-3, and PARP, induces apoptosis in a caspase-dependent manner and inhibits the NF-κB and PI3K/Akt/mTOR signaling pathways.
AE-848 (12.5 mg/kg, i.p.) significantly inhibits tumor growth and prolongs overall survival of MM cell-bearing mice.
