ADT-094 (ADT094) is a novel, non-COX inhibitory sulindac derivative that potently and selectivity inhibits colon tumor cell growth by inhibiting PDE5 and PDE10 with IC50 of 2.4 uM and 0.5 uM, respectively.
ADT-094 displays no inhibitory activity for COX-1 and COX-2.
ADT-094 activates cGMP/PKG signaling and suppresses β-catenin/TCF-transcriptional activity, resulting in cell cycle arrest and apoptosis induction.
ADT-094 inhibits the growth of human HCT116 and HT29 colon tumor cell lines (IC50=1 uM).
