AB-343 is a potent, selective, reversible covalent inhibitor of SARS-CoV-2 Mpro with Ki/IC50 of 2.8/8 nM, inhibits SARS-CoV-2 in Vero E6 cells and SARS-CoV-2-infected human lung cells (hACE2-A549) with EC50 of 18 nM.
AB-343 shows no inhibition against a panel of human proteases and other viral proteases (IC50 >30 μM), except against caspase 2 and cathepsin K.
AB-343 potently inhibits Mpro of other human betacoronaviruses (SARS-CoV, MERS-CoV, hCoV-OC43, and hCoV-HKU1) and alphacoronaviruses (hCoV-NL63 and hCoV-229E) with IC50 values ranging from 11 to 104 nM and Ki values ranging from 5.3 to 45 nM.
AB-343 exhibits antiviral activity against MERS-CoV in Vero E6, and hCoV-OC43 in both Huh7 and MRC5 cells.
AB-343 shows high potency against Mpro from SARS-CoV-2 variants of concern.
