A4276 is a novel orally bioavailable NAMPT inhibitor with IC50 of 492 nM for inhibiting NAMPT enzymatic activity, shows high selectivity for NAPRT-deficient lung cancer cell lines.
A4276 exhibits remarkable selectivity against NAPRT-deficient EMT-like cancer cell lines across various cancer types while effectively sparing NAPRT-positive cells, particularly, normal cells.
A4276 (100 mg/kg) potently inhibits the growth of NAPRT-negative EMT-subtype cancer in in HGC27 xenografts. A4276 delays Wallerian degeneration, prevents chemotherapy-induced peripheral neuropathy (CIPN).
