A-867744 is a potent, selective α7 nAChR positive allosteric modulator (PAM) with Ki of 23 nM, potentiates acetylcholine (ACh)-evoked currents with EC50 of 1 uM; A-867744 failed to potentiate responses mediated by 5-HT3A or α3β4 and α4β2 nAChRs; increases choline-evoked α7 currents and recovery from inhibition/desensitization, and enhances spontaneous inhibitory postsynaptic current activity at 10 uM in rat hippocampus; normalizes gating deficits in rodent model of sensory gating.
