2-MPPA (GPI-5693) is a potent, selective and orally active glutamate carboxypeptidase II (GCP II, NAALADase) inhibitor with IC50 of 90 nM.
2-MPPA suppresses mossy fiber-CA3 synaptic neurotransmission.
2-MPPA attenuates morphine tolerance but not dependence in C57/Bl mice.
2-MPPA is effective in preclinical models of diseases where excess glutamate release is implicated, including neuropathic pain.
