(S)-4CMTB (S-4CMTB, AMG 7703, Phenylacetamide 1) is a potent, specific and allosteric agonist of FFA2 (GPR43) with cAMP IC50 of 0.7 and 0.96 uM for hFFA2 and mFFA2, respectively.

Phenylacetamide 1 is inactive against a panel of GPCRs that included FFA1, FFA3, GPR109A, growth hormone secretagogue receptor, endothelin type B receptor, CCR2, CXCR3, CXCR4, and CCR7 at 30 uM.

Phenylacetamide 1 is capable of activating both Gαq- and Gαi -coupled pathways for receptor function.

Phenylacetamide 1 inhibits lipolysis in 3T3L1 adipocytes in a concentration-dependent manner as measured by the reduction of glycerol production in adipocytes.
