(R)-HTS-3 is a small-molecule inhibitor of lysophosphatidylcholine acyltransferase 3 (LPCAT3) with IC50 of 0.09 uM, an integral membrane acyltransferase in the Lands cycle responsible for generating C20:4 phospholipids.
(R)-HTS-3 inhibits LPCAT3 in a biphasic manner, possibly reflecting differential activity at each subunit of the LPCAT3 homodimer.
(R)-HTS-3 treatment cause rapid rewiring of polyunsaturated phospholipids in human cells.
LPCAT3 has been implicated in key biological processes such as intestinal lipid absorption, lipoprotein assembly, and ferroptosis.
