Z640 is a novel non-selective atypical PKC (aPKC) agonist, increases PKCζ and PKCι kinase activity with EC50 of 1.09 and 3.47 µM in vitro kinase activity assays, respectively.
Z640 binds to PKCι in vitro with an affinity value of 1.37 uM.
Z640 can protect neuronal cell lines against Aβ oligomer-induced toxicity (EC50 of 4.57 µM in WT7 cells).
Z640 (10 uM) can protect cells by correcting Aβ oligomer-induced ROS elevation and mitochondria damage in N2a cells, also reduces Aβ oligomer-induced ROS elevations in a dose-dependent manner (EC50=5.11 µM).
Z640 can regulate amyloid precursor protein processing and effectively reduces Aβ40 generation with EC50 of 2.73 uM.
