SIC-19 is a potent, selective SIK2 inhibitor, promotes SIK2 protein degradation via the ubiquitination pathway, exhibits synthetic lethality with PARP inhibitors in ovarian cancer.
potently inhibits six ovarian cancer cell lines with IC50 values ranging from 2.13 μM to 9.74 μM.
SIC-19 significantly enhances response to PARP inhibitors in ovarian cancer cell lines, ovarian cancer organoids and xenograft models.
SIC-19 decreases the phosphorylation of RAD50 and blocks the nuclear translocation of RAD50, impaires nuclear filament assembly and DNA repair.
SIC-19 alone significantly inhibited tumor growth in SKOV3 xenografts, the combination of SIC-19 and olaparib demonstrates even greater inhibition of tumor growth than either single agent alone.
