Non-canonical p38α inhibitor NC-37 (NC-p38i NC-37) is a potent, highly selective, non-canonical inhibitor of p38α, inhibits TAB1-induced p38α autophosphorylation in vitro with IC50 of 0.27 uM.
NC-37 (1-30 uM) significantly decreases TAB1-induced p38α phosphorylation levels in a dose-dependent manner in HEK293T cells.
NC-37 does not affect the canonical activation of p38α by the MAP2K MKK6, showed limited activity agianst the kinase activity of p38α on the substrates MK2 and ATF2 at 10 uM.
NC-37 binds to and stabilizes p38α without inhibiting its interaction with TAB1.
NC-37 effectively inhibits p38α autophosphorylation by weakly binding to several regions in and around the ATP-binding pocket of p38α.
