NBM-T-L-BMX-OS01 (BMX, HDAC8 inhibitor) is a potent, selective and BB penetrant HDAC8 inhibitor with IC50 of 831 nM.
BMX displays >4,000-fold selectivity over HDAC3.
BMX shows less neurotoxicity than SAHA on primary hippocampal neuron.
BMX (5 mg/kg/d, i.p.) enhances spatial learning on water-maze memory acquisition and retrieval in rats.
BMX (5 mg/kg/d, i.p.) enhances passive one-way avoidance tasks on passive one-way inhibitory avoidance learning in rats.
BMX ameliorates scopolamine-(Sco-) induced learning and memory impairment in animals.
BMX shows significant anti-cell proliferation effects in CRC cells, human umbilical endothelial cells, lung cancer cells, and glioblastoma cells, and it also exhibits tumor suppression ability in an animal xenograft model.
Combination of BMX and temozolomide (TMZ) triggers cell cycle arrest, senescence, autophagy, and apoptosis in CRC cells via upregulation of p53/p21/E2F3/Bax.
