MW181 (MW01-10-181SRM) is an isoform-selective, brain-permeable, and orally bioavailable small molecule inhibitor of p38α MAPK with Ki of 184 nM, no affitnity for CK1δ.
MW181 suppresses p38α MAPK activation, reduces tau phosphorylation, and decreases proinflammatory cytokine production.
MW181 suppresses LPS-induced increases in mouse brain IL-1β, TNFα, IL-6, IL-10, and CXCL1 in mice.
MW181 significantly increases synaptophysin protein expression and prevents cognitive impairment in the aged hTau mice.
