HOSU-53 sodium is selective and potent dihydroorotate dehydrogenase (DHODH) inhibitor with IC50 of 0.95 nM in cell-free human DHODH enzyme inhibition assays.
HOSU-53 shows relatively similar inhibitory potency across mouse, rat, and dog DHODH enzyme.
HOSU-53 displays little to no activity against a panel of approximately 450 human kinases at 10 μM.
HOSU-53 exhibits potent in vitro antileukemic activity with median IC50 of 120.5 nM, promotes in vitro innate phagocytic function.
HOSU-53 (30 mg/kg) demonstrates in vivo single-agent antileukemic activity, results in superior in vivo outcome using AML disseminated xenograft model.
HOSU-53 demonstrates in vivo synergy with approved targeted AML therapies.
