HEC-73543 is a novel potent, selective, orally bioavailable FLT3 inhibitor for the treatment of refractory acute myeloid leukemia (AML); shows proliferation inhibition against MV-4-11 and MOLM-13 cells with IC50 of 1.5 and 1.4 nM, respectively; potently inhibits the phosphorylation of FLT3 and its downstream signaling kinases STAT5, AKT and ERK1/2 in tumor tissues, exhibits antitumor activity in an FLT3-ITD subcutaneous tumor xenograft model.
